Giakountis et al. identify a Wnt-regulated long non-coding RNA in colorectal cancer cells that positively regulates the expression of its genomic neighbor ASCL2, a transcription factor that controls intestinal stem cell fate. It does so through orchestrating long-distance looping, factor recruitment and associated chromatin changes at the affected genomic loci. The WiNTRLINC1-ASCL2 regulatory axis is amplified in colorectal cancer.
